Lords of Thunder is a fantasy-themed scrolling shoot 'em up game in which the player plays the knight Landis, wearing the armor of his ancestor Drak, against Zaggart of Garuda Empire, who resurrected the evil deity Deoric, and his six dark generals across Mistral. At the game's title screen, a configuration menu can be opened before starting. Each difficulty setting arranges stage dangers and enemy patterns. The configuration screen has a cheat code for a hidden boss rush mode.

When starting the game, the player can choose any of the first six stages, similar to Thunder Force III, with the final stage unlocked after all six are cleared. Once the stage is picked, the player chooses one of four armors (based on the four classical elements) to use, influencing Landis' shoots and explosives and the difficulty. The player can buy Landis things with crystal currency before a stage begins. Extra bombs, shields, and health-restoring elixirs. The player controls Landis over a constantly scrolling background filled with enemies and obstacles until a boss is found, which must be beaten to clear a stage and move on.
